Abstract
OBJECTIVE To establish a headspace capillary gas chromatography for determining the residual organic solvents acetone and thioglycol in coenzyme A. METHODS Using a HP-INNOWAX capillary column (30 m×0.53 mm×1 μm), flame ionization detector (FID) and adopting headspace injection. RESULTS The calibration curves of each solvent had good linear relationship(r>0.999), The average recoveries was 101.29% (RSD=1.32%, n=9), 98.67% (RSD=1.71%, n=9), and the detection limit was 2.52 μg·mL-1, 3.95 μg·mL-1. The content of acetone ranged from 0.06% to 4.33%,and the content of thioglycol ranged from 0.14% to 0.72%. CONCLUSION The proposed method offers simple and fast operation, high sensitivity and accuracy, and it is a substitute method to determinate the residual organic solvents in coenzyme A.
