Abstract
OBJECTIVE To extract polysaccharides from the root of Taraxacum and then isolate, purify and analyze the structure of polysaccharides. METHODS Polysaccharides were extracted by ultrasonic and coordinated enzyme methods, after separating and purifying, four levels (A, B, C, D) were fractionated by Sephadex G-75 column and then monosaccharide constituents were analyzed by HPLC applied to PMP precolumn derivation. Preliminary analysis of structure was analyzed by UV, IR method. RESULTS All of the four kinds of monosaccharides contain D-rhamnose, glucose, D-galactose, D-xylose and D-arabinose. Their molar ratios are as follow in turns: Fraction A: 0.098∶2.88∶0.125∶0.017∶0.33;B: 0.113∶0.68∶0.13∶ 0.011∶0.19; C: 0.139∶2.46∶0.36∶0.13∶0.086; D: 0.028∶0.36∶0.031∶0.012∶0.94. CONCLUSION It showed that there were significant differences in four species of polysaccharides.
