Abstract
OBJECTIVE To extract polysaccharide from Fomes officinalis Ames of Xinjiang and evaluate its immunity activity in vitro. METHODS Crude polysaccharide was extracted with hot water, precipitated with 95% ethanol, removed protein with Sevag method. Polysaccharide content was determined with phenol-sulfuric acid method. It was identified by IR and UV scanning spectrum. MTT method was applied to determine the immunity activities of the polysaccharide from Fomes officinalis Ames(FoP). RESULTS The yield of crude water-extracted polysaccharide was 0.85%. The polysaccharide content was 68.81%. IR spectrum indicated that there were typical characteristic absorption peaks of polysaccharide. UV scanning spectrum showed that there were no absorption peaks of protein and nucleic acid at point 280 nm and 260 nm. FoP increased the proliferation of mice spleen lymphocytes and peritoneal macrophages cell populations in vitro. CONCLUSION FoP has certain immunity activity.
