Comparison between HPLC and Microbiological Method for Determination of the Content of Vancomycin in Human Serum

OBJECTIVE To compare HPLC and microbiological assay for determination of the content of vancomycin in human serum. METHODS Thirty patients were given 1.0 g vancomycin by infusion，q.d..The serum concentration were determined by HPLC and bioassay，respectively. RESULTS The calibration curve of HPLC was within the range of 5.0-80.0 μg·mL-1. The recovery of HPLC method was 97.3% to 101.6%，within-day and day-to-day RSD were 1.74%-4.06%，and 1.22% -4.21%, respectively. The calibration curve of microbiological assay was linear in the range of 5.0-60.0 μg·mL-1. The recovery of microbiological assay was 98.2%-100.8%，within-day and day-to-day RSD were 4.22%-5.51% and 4.59%-6.49%，respectively. CONCLUSION The recovery and precision of the both methods are similar. There were no significant differences.
