Studies on the Hypoglycemic Activity of 1,2,3-inositol Triphosphate in Diabetes Mellitus Mice Induced by Alloxan

Abstract
OBJECTIVE To investigate the effects of 1,2,3-inositol triphosphate Ins(1,2,3)P3 on the blood glucose of Alloxan-diabetic mice. METHODS Phytic acid was partially hydrolyzed by phytase from wheat bran. The partial hydrolyzed solutions were pumped into a column containing 717 strong anion-exchange resin, then delivered a linear gradient of 0.05-0.7 mol·L-1 HCl giving Ins(1,2,3)P3. Alloxan-induced diabetic mice were divided into 5 groups randomly, and intra-peritoneal injected with saline,10, 50, 100 mg·kg-1·d-1 Ins(1,2,3)P3, 70 mg·kg-1·d-1 PHF respectively for 12 days successively. Fasting orbital blood samples were collected for measurement of glucose at the end of the experiment. RESULTS Ins(1,2,3)P3 was found to reduce blood glucose level in alloxan-diabetic mice markedly at doses of 50,100 mg·kg-1·d-1, but no effect on normal mice. Compared with the model control group, Ins(1,2,3)P3 had the ability to raise the thymus and spleen indexes of diabetic mellitus mice and normal mice,inhibiting the atrophy of thymus and pancreas of the diabetic mice induced by alloxan. CONCLUSION Ins(1,2,3)P3 has good hypoglycemic effect on Alloxan-diabetic mice.
