OBJECTIVE To explore the pharmacological effects of Fupo Ganmao Granules against influenza virus and secondary bacterial infections, and to provide experimental evidence for its clinical application.
METHODS To detect the inhibitory rates of Fupo Ganmao Granules on influenza viruses H1N1, H3N2 and FluB through in vitro experiments; to establish mouse pneumonia models infected with H1N1 and FluB, and observe the effects of Fupo Ganmao Granules on lung coefficient, relative viral load in the lungs, serum inflammatory factor levels and pulmonary histopathology; to establish a mouse model of H1N1 secondary bacterial(Klebsiella pneumoniae) infection, and observe the effects of Fupo Ganmao Granules on the mortality rate and life extension rate of mice.
RESULTS The therapeutic indices of Fupo Ganmao Granules in the in vitro inhibitory experiments on H1N1, H3N2 and FluB viruses were 2.4, 2.1 and 2.7 respectively. In vivo experiments showed that Fupu Ganmao Granules reduced the relative viral load in the lungs and ameliorated lung lesions in mice with H1N1 and influenza B (FluB) pneumonia; decreased serum levels of the inflammatory factors TNF-α, IFN-γ, IL-1, IL-6, IgG, IgM, and CCL2(P<0.05 or P<0.01), and reduced the lung index in mice with FluB pneumonia(P<0.05). Moreover, it could reduce the mortality rate and prolong the survival time of mice with virus-induced secondary bacterial infection(P<0.05 or P<0.01).
CONCLUSION Fupo Ganmao Granules exhibit good efficacy in anti-influenza virus activity and treating secondary bacterial infections, which may serve as an important basis for its treatment of influenza and related complications.
