Adverse Reactions and Pharmaceutical Care of Antibody-drug Conjugates

    • Antibody-drug conjugates(ADCs) are novel tumor therapeutics composed of a linker, cytotoxic drug and monoclonal antibody. Compared with traditional treatments such as surgery, radiotherapy, and chemotherapy, ADCs significantly reduce toxicity to normal tissues, while retaining the potent tumor cell-killing effect. Since the first ADCs was approved in 2000, ADCs have been widely used in various tumors, including hematologic neoplasms and solid tumors. However, the adverse reaction, involving hematological, peripheral nerves, lungs, liver and other organs, cannot be ignored. Thus, this paper comprehensively review the structural composition, mechanism of action, adverse reactions and pharmaceutical care of ADCs, aiming to provide references and suggestions for the rational clinical application of ADCs.
