OBJECTIVE To analyze the main components of Baidoukou aqueous extract and the components that enter the blood of rats after oral administration by UPLC-Q-Exactive-Orbitrap MS/MS technology.
METHODS Using ACQUITY UPLC HSS T3 (2.1×100 mm, 1.8 μm) column; Using A(deionized water, containing 0.1% formic acid) and B (acetonitrile, containing 0.1% formic acid) as mobile phases, gradient elution, column temperature: 35°C; injection volume: 10 μL; flow rate: 0.2 mL·min−1. Using electrospray ionization source (ESI), detect with Full ms/dd-ms2 mode.
RESULTS A total of 70 components were identified from the aqueous extract of Baidoukou, including organic acids, flavonoids, amino acids, alkaloids, and lactones. 19 prototype components were identified in the drug-containing serum samples, including 10 organic acids such as artemisinic acid and azelaic acid, 2 lactones such as isoalantolactone and dehydrocostus lactone, 2 alkaloids such as Betaine and Xanthine, 3 amino acids such as L-Tyrosine and L-Tryptophan.
CONCLUSION It’s sclarified for the first time the chemical composition of Baidoukou aqueous extract and its oral blood components in this study, providing a theoretical basis for its further research and development.
