    Simultaneous Determination of Five flavones in Polygonum perfoliatum L. and Its honey processed products by HPLC

    • OBJECTIVE to establish an HPLC method for the determination of hyperoside, kaempferol-3-o-rutin, quercetin, kaempferol and isorhamnetin in Polygonum perfoliatum L. and its honey processed products. METHODS The analysis was performed on Phenomonex®-C18 column(250 mm×4.6 mm, 5 μm) with the gradient elution of methanol(A)-0.4% phosphoric acid(B) solution at flow rate of 1.0 ml·min-1. The column temperature was 30℃ and the detection wavelength was set at 360 nm. RESULTS The linear ranges of hyperoside, kaempferol-3-o-rutin, quercetin, kaempferol and isorhamnetin were 3.406-340.6, 0.828-82.8, 5.025-502.5, 1.036-103.6, 1.162-116.2 μg·mL-1, respectively (r ≥ 0.999 3), the average recoveries of five components were 100.04%, 99.67%, 97.96%, 102.50% and 98.99%, respectively. After honey precessed, contents of hyperoside and kaempferol-3-O-rutoside in Polygonum perfoliatum decreased significantly, while the contents of quercetin, kaempferol and isorhamnetin increased significantly. CONCLUSION The method is simple, efficient, accurate and reliable, which can provide scientific basis for the quality control of the Polygonum perfoliatum L. and its honey processed products.
