Abstract
OBJECTIVE To establish a HPLC method for the simultaneous determination of morphine, codeine phosphate and bergenin in Keqing capsules. METHODS Ultimate®AQ-C18(4.6 mm×250 mm, 5 µm) column was used; mobile phase was acetonitrile(A)-0.1% phosphoric acid(B) with gradient elution; flow rate was 1.0 mL·min–1; column temperature was 35 ℃; detection wavelength was 210 nm and the injection volume was 10 µL. RESULTS Good linear relationship was observed for morphine, codeine phosphate and bergenin in the range of 20.018 2-320.291 2 µg·mL–1 (r=0.999 9), 4.031 0-64.496 6 µg·mL–1 (r=1.000 0), and 19.760 0-316.160 0 µg·mL–1(r=1.000 0), respectively. The average recoveries(n=6) were 98.33%(RSD=2.72%), 98.72%(RSD=2.77%) and 98.54%(RSD=1.06%), respectively. CONCLUSION The established HPLC method is simple, accurate, repeatable and can be used for the determination of morphine, codeine phosphate and bergenin in Keqing capsule. It can also provide reference for its quality control.
