    Effect of Meglumine Cyclic Adenylate on Proliferation, Migration, Angiogenesis and Apoptosis in Human Umbilical Vein Endothelial Cells

    • OBJECTIVE To explore the effects of meglumine cyclic adenylate(MCA) on the proliferation, migration, lumen formation and apoptosis of human umbilical vein endothelial cells(HUVEC). METHODS The effects of different concentrations of MCA on HUVEC cell morphology, proliferation, migration, luminal formation ability and apoptosis-related protein expression were observed by microscope observation, MTT method, scratch repair experiment, lumen formation experiment and Western blotting. RESULTS Microscopic observation revealed that with the increase of the dose of MCA, the morphology of HUVEC cells changed from polygonal to oval. MCA could inhibit HUVEC proliferation, migration and lumen formation in a concentration-dependent manner, and high-dose MCA could also induce cell apoptosis, which was manifested by an increase in the number of apoptosis, the expression of Bax, Cleaved-PARP protein was up-regulated and the expression of PARP and Bcl-2 protein was down-regulated. CONCLUSION Meglumine cyclic adenylate can inhibit HUVEC cell proliferation, migration, luminal formation and induce cell apoptosis.
