Study on Purification Process and Anti-inflammatory Activity of Total Iridoid Glycosides from Patrinia Scabra Bunge

Abstract
OBJECTIVE To optimize the technological conditions for the enrichment and purification of total iridoid glycosides from Patrinia scabra Bunge by macroporous resin and study its anti-inflammatory activity. METHODS The content of total iridoid glycosides was determined through the ultraviolet spectrophotometry and was taken as an index. The macroporous resin which was suitable for purifying the total iridoid glycosides from Patrinia scabra Bunge was screened through the static and dynamic adsorption-desorption ability. Furthermore, the enrichment and purification process of Patrinia scabra was optimized by investigating the sample concentration, optimum sample volume, ethanol volume fraction, elution dosage, elution flow rate and so on. The anti-inflammatory activity of the total iridoid glycosides from Patrinia scabra Bunge was evaluated by the xylene-induced ear swelling test with the aspirin as positive control group. RESULTS AB-8 macroporous resin was the optimal material for the purification of total iridoid glycosides from Patrinia scabra Bunge. The optimum purification conditions were as follows:the dosage of sample volume was 4 BV, the concentration was 1.10 mg·mL-1, the dynamic adsorption was carried out at the flow rate of 2 BV·h-1, the impurity was washed out with 4 BV water, and then the desorption was carried out with 50% ethanol of 4 BV at the flow rate of 2 BV·h-1. Under these conditions, the content of total iridoid glycosides increased from 8.27% to 35.92% after purification, which was about 4.4 times of that before purification. The high-dose group of total iridoid glycosides(200 mg·kg-1) could effectively reduce the xylene-induced ear swelling in mice, and the inhibition rate reached 53.20%, which was significantly compared with the blank control group. CONCLUSION The optimized purification process with AB-8 macroporous resin as filler is feasible, and the total iridoid glycosides of Patrinia scabra Bunge possess certain anti-inflammatory effect.
