Effects of Different Polar Elution Parts of Vinegar Bupleuri Radix on the Distribution of Baicalin in Mice and Its Component Analysis

    • OBJECTIVE To investigate the effect of different polar elution parts of vinegar Bupleuri Radix on the distribution of baicalin in mice, and to preliminary explore its synergistic active components in different organs of vigenar Bupleuri Radix combined with baicalin by correlation analysis. METHODS Study on the effects of distribution of baicalin of different polar elution parts of vinegar Bupleuri Radix by multiple doses; the content of baicalin in each tissue was determined by LC-MS/MS; Q-Exactive Plus was used to analyze the components of different polar elution parts of vinegar Bupleuri Radix and the possible active components, and the correlation analysis was carried out by SPSS 21.0 software to explore the possible active components. RESULTS Compared with single group of baicalin, the combined group(95% ethanol elution part) increased the concentration of baicalin in liver, lung and spleen, 75% ethanol elution part increased the concentration of baicalin in lung, and 50% ethanol elution part increased the concentration of baicalin in spleen. Correlation analysis showed that 21 compounds in vinegar Bupleuri Radix were significantly positively correlated with the distribution of baicalin in liver, mainly acetylated saponins produced by vinegar preparation. The other 13 compounds showed significant correlation with the distribution of baicalin in the lung, most of them were raw saikosaponin. CONCLUSION Acetylated saponins, primary saponins may separately be the active components in the treatment of liver and lung, the results need to be further verified.
