Analysis of Cyclocarya Paliurus Leaves by UPLC-Q-TOF-MS and Its Effect on Testicular Restoration

Abstract
OBJECTIVE To analyze the chemical composition of the water extract of Cyclocarya paliurus leaves and its influence on testicular tissue in diabetic mice. METHODS The water extract of Cyclocarya paliurus leaves was analyzed by ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS), and the chemical identification was then carried out based on the high-resolution molecular weight and fragment ion, as well as mass spectral database searching, the α-glucosidase inhibitory activity of some compounds was determined by PNPG method. Therapeutic effect of the Cyclocarya paliurus leaves on alloxan-induced diabetic mice was evaluated at doses of high(2 000 mg·kg-1·d-1), medium(1 000 mg·kg-1·d-1) and low(500 mg·kg-1·d-1). RESULTS Twenty compounds were identified, and out of them, morinIC50=(3.98±1.38)μg·mL-1, umbelliferoneIC50=(33.48±2.54)μg·mL-1, and methyleugenolIC50=(34.63±4.66)μg·mL-1 were first identified as α-glucosidase inhibitors from Cyclocarya paliurus leaves. There were significant different in body weight and blood glucose between the high, medium and low dose groups compared with the model group, and section diagram of testicular tissue showed that the high dose group has a better repair effect than the positive group. CONCLUSION The water extract of Cyclocarya paliurus leaves has significant hypoglycemic effect and good repair function to testicular tissue.
