    Effect of Zingiberis Rhizoma Recens on Dissolution Rate of Chemical Components in Scutellariae Radix Under Different Compatibility Ratios

    • OBJECTIVE To reveal the scientific connotation of the compatibility of Zingiberis Rhizoma Recens and Scutellariae Radix based on the microscopic quantitative standard, and provide some scientific references for clinical application, the change of chemical components dissolution rate of Scutellariae Radix was explored before and after different proportion of Scutellariae Radix was mixed with Zingiberis Rhizoma Recens. METHODS The separation was performed on Agilent C18 column(4.6 mm×150 mm, 5 μm) with mobile phase composed of methanol-0.1% phosphoric acid solution for gradient elution at the flow rate of 1 mL·min-1, the column temperature was set at 30℃, the injection volume was 20 μL, and the detection wavelength was set at 280 nm. HPLC chromatogram of extracts with different proportions of Scutellariae Radix and Zingiberis Rhizoma Recens was established, and the common peaks were demarcated with the single chromatogram of Scutellariae Radix as the control. The changes of each chromatogram under different compatibility ratios were analyzed, and the relationship between the relative peak area of characteristic peaks of Scutellariae Radix and compatibility ratio was investigated, and the content changes of four index components in Scutellariae Radix were analyzed. RESULTS Thirty chromatographic peaks were isolated from single extract of Scutellariae Radix, and the peaks of 15, 19, 24 and 29 corresponded to baicalin, wogonside, baicalein and wogonin respectively. Twenty-two common peaks were marked in the chromatograms under different compatibility ratios. Compared with single Scutellariae Radix, the relative peak area of characteristic peaks of Scutellariae Radix decreased in different degrees in the rest proportion except individual chromatographic peaks, but the decrease degree was the smallest in 1:1 ratio. In this ratio, the content of wogonin, baicalein and wogonin besides baicalin was not significantly lower than that of single product, while the content of the above four components in the other ratios was significantly lower(P<0.05). CONCLUSION The dissolution rate of chemical components in Scutellariae Radix can be reduced when it is mixed with Zingiberis Rhizoma Recens in different proportions, but not all components are reduced. The compatibility of Scutellariae Radix and Zingiberis Rhizoma Recens 1:1 has the least influence on the dissolution rate of Scutellariae Radix.
