Effect of Wumoyin Decoction on Regulating Brain-gut Peptide in Functional Dyspepsia Rats with Spleen Deficiency and Qi Stagnation Syndrome

Abstract
OBJECTIVE To study the effect of Wumoyin decoction on regulating functional dyspepsia rats with spleen deficiency and qi stagnation syndrome, explore its mechanism of action. METHODS Except the normal group, the other rats were modeled by "chronic restraint+swimming+irregular eating" compound factor method. The rats were randomly divided into model group, domperidone group, Chaishao Liujunzi decoction group, Wumoyin decoction high dose group(6.2 g·kg-1), middle dose group(3.1 g·kg-1) and low dose group(1.55 g·kg-1). Besides the normal group and model group, the other groups of rats were given the corresponding drug intervention for 14 d. The content of Ghrelin, MTL, GAS, CCK, VIP, CGRP in plasma were detected by ELISA method, and the expression of Ghrelin, MTL, GAS, CCK, VIP, CGRP in gastric antrum tissue and hypothalamus were detected by immunohistochemistry method. RESULTS Compared with the model group, the Ghrelin and MTL in plasma of rats in Wumoyin decoction each dose group was significantly raised(P<0.01), and the CCK was very significantly reduced(P<0.01). The VIP in plasma of rats in the high dose and middle dose groups were significantly reduced(P<0.05). The CGRP in plasma of rats in the high dose group was significantly reduced(P<0.05). There was no significant difference in the expression of Ghrelin, MTL, GAS, CCK, VIP and CGRP in gastric antrum of rats in each dose group. The expression of Ghrelin in the hypothalamus of the rats in the high dose group was significantly increased(P<0.01). The expressions of Ghrelin, MTL, GAS, VIP, CGRP in the hypothalamus of rats in the middle dose group were significantly increased(P<0.01). CONCLUSION Wumoyin decoction has the effect of regulating the expression of brain gut peptide in rats with functional dyspepsia of spleen deficiency and qi stagnation syndrome, which may be one of its mechanisms.
