    Determination of Vancomycin Concentration in Synovial Fluid of Patients with Prosthetic Joint Infection by HPLC

    • OBJECTIVE To establish a method for determination of vancomycin concentration in synovial fluid of patients with prosthetic joint infection by HPLC. METHODS Used tinidazole as internal standard substance, vancomycin concentration in synovial fluid was analyzed by Hypersil C18(250 mm×4.6 mm, 5 μm) with mobile phase of acetonitrile-0.012 5 mol·L-1 potassium dihydrogen phosphate buffer solution(pH=3.2)(10:90). The flow rate was 1 mL·min-1. The column temperature was 35℃ and the injection volume was 10 μL. RESULTS Vancomycin concentration in synovial fluid was in well linearity in the range of 0.906 8-90.681 8 μg·mL-1. The relative standard deviations of intra-batch and inter-batch precision was <15%, and the accuracy was between 85.89%-103.51%. CONCLUSION The method is simple, sensitive and accurate, which is suitable for monitoring of vancomycin concentration in synovial fluid of patients with prosthetic joint infection.
