Effects of PMIEE on Proliferation and Apoptosis of HepG2 and MCF7 cells

Abstract
OBJECTIVE To investigate the effects of Patrinia monoterpene iridoid ether esters (PMIEE) on the proliferation and apoptosis of HepG2 and MCF7 cells. METHODS The HepG2 and MCF7 cells were cultered in vitro. CCK8 assay was used to detect cell proliferation inhibition and Annexin V-FITC/PI double labeling flow cytometry was used to detect cell apoptosis and cell cycle, and cell scratch assay was used to detect cell migration. Detection of the expression of Bcl-2, Bax, caspase-3, cdc2 and CyclinB1 by Western blot. RESULTS CCK8, scratch and Annexin V-FITC/PI flow cytometry showed that PMIEE significantly inhibited the proliferation of HepG2 and MCF7 cells, and promoted apoptosis and decreased mobility (P<0.05), showed a dose-effect relationship. The cycle arrest of HepG2 cells by PMIEE was mainly in G2/M phase, while MCF7 cells was mainly in G0/G phase. The results of Western blot showed that PMIEE could significantly down-regulate the expression of Bcl-2, cdc2, CyclinB1 and up-regulate the expression of Bax and caspase3 in two kinds of cells. CONCLUSION PMIEE can induce proliferation inhibition and apoptosis of HepG2 and MCF7 cells, down-regulate the expression of Bcl-2, cdc2 and CyclinB1, and up-regulate the expression of Bax and caspase3. While its potential related with anti-cancer mechanism.
