Determination of Contents of Dihydromyricetin, Rutin and Myricetin in Ampelopsis Grossedentata Stems by HPLC

Abstract
OBJECTIVE To establish a method for the determination of contents of flavonoids and analyze the contents of dihydromyricetin, rutin and myricetin in Ampelopsis grossedentata stems from different areas of Guangxi. METHODS HPLC method was adopted. The separation was performed on Agilent PoroshellEC-C18(4.6 mm×250 mm, 4.0 μm) column and the mobile phase consisted of acetonitrile and 0.1% H3PO4 water solution in gradient elution at flow rate of 1 mL·min-1, the column temperature was 25, the detection wavelengths were set at 330℃ nm for dihydromyricetin and 375 nm for myricetin. RESULTS The linear ranges were 0.800 5-9.606 μg(r=0.999 9) for dihydromyricetin, 0.201 6-2.419 2 μg(r=0.999 8) for rutin and 0.100-1.202 μg(r=0.999 7) for myricetin, and the average recovery were 98.1%, 101% and 99.9%, respectively, RSDs<3%. The determined results showed that the contents of dihydromyricetin, rutin and myricetin in Ampelopsis grossedentata stems from Lingchuan of Guilin were higher, and the contents in dihydromyricetin and myricetin in Ampelopsis grossedentata stems from Shuangjiang of Wuming and Huzhu Cun of Jinxiu were lower; the contents of rutin in Ampelopsis grossedentata stems from Shuangjiang of Wuming were higher, and the contents of rutin from Siding Zhen of Liuzhou were lower. CONCLUSION The contents of dihydromyricetin, rutin and myricetin in Ampelopsis grossedentata stems from different areas were quite different. This method can be better to separate from coexisting component, with good separation, and the results showed the method had high accuracy and good reproducibility.
