Abstract
OBJECTIVE To explore the protective effect and mechanism of Qingnao tablet on cerebral ischemia reperfusion in gerbils. METHODS Gerbil's bilateral carotid arteries were occluded for 10 min and then reperfused 60 min to make cerebral ischemia gerbil model. The effects of large, medium and small dose of Qingnao tablet group(1.5, 0.75, 0.375 g·kg-1) on interleukin-1β(IL-1β), tumor necrosis factor alpha (TNF-α), excitatory amino acid glutamate (Glu), aspartic acid (Asp), thromboxane B2(TXB2), malondialdehyde (MDA) content and the activity of ATP and immunoreactive expression of NF-κB in gerbils were observed. RESULTS Compared with model group, IL-1, TNF-α, Glu, Asp, TXB2 brain tissue, the content of MDA and NF-κB positive expression were significantly decreased by large and medium dose of Qingnao tablet (P<0.01 or P<0.05); Then the activity of brain tissue Na+-K+-ATP, Mg2+-ATP Ca2+-ATP, Ca2+-Mg2+-ATP were significantly increased (P<0.01 or P<0.05). CONCLUSION Qingnao tablet on gerbil cerebral ischemia reperfusion injury has obvious protective effect, its mechanism may be related to the inhibition of peroxidation of inflammatory cytokines, and improve enzyme activity.
