Effects of Folium Mori on Serum Uric Acid Levels and Gut Microbiota in Rats with Hyperuricemia

Abstract
OBJECTIVE To observe the effects of Folium mori Macha (FM) on blood uric acid level and gut microbiota regulation. METHODS Hyperuricemia model was established by feeding SD rats with high-purine diet, rats in experiment groups were orally administered FM at the same time. At the end of experiment, serum levels of uric acid (UA), creatinine (Cr), blood urea nitrogen (BUN) were determined by automatic biochemical analyser and xanthine oxidase (XOD) activities in serum and liver tissue and serum lipopolysaccharide (LPS) levels were determined by ELISA according to the specification of the kits; Faeces was collected and DNA of intestinal microbiota was exeracted then gut microbiota composition was evaluated by 16S rDNA sequencing. RESULTS FM could significantly decrease serum UA, Cr, BUN levels and inhibit XOD activities in both serum and liver tissue. It could increase the ratio of Lactobacillus in intestinal floras and decrease serum LPS levels. RAD analysis showed that the operational taxonomic unit (OTU) level of intestinal floras was related to the level of serum UA and the activities of XOD. CONCLUSION FM possess a function on anti hyperuricemia via regulating intestinal floras, decreasing serum LPS level and inhibiting XOD activities.
