Abstract
OBJECTIVE To develop a method of quantitative analysis of multi-components by single marker(QAMS) for determination of chlorogenic acid, emodin, chrysophanol, dl-tetrahydropalmatine in Fule granula. METHODS The Agilent ZORBAX Eclipse XDB C18(4.6 mm×250 mm, 5 μm) column was used; mobile phase was acetonitrile (A)-0.2% phosphoric acid solution(B). gradient elution:0-8.0 min, 85% B; 8.0-15.0 min, 85%→45% B; and 15.0-30.0 min, 45%→20% B. Wavelengths:0-8.0 min was 327 nm, 8.0-15.0 min was 254 nm and 15.0-30.0 min was 280 nm. Flow rate was 1.0 ml·min-1 with column temperature 30℃. Relative correction factors of chlorogenic acid, chrysophanol and tetrahydropalmatine were determined with emodin as internal standard. The method was evaluated for reproducibility, and the difference between calculated and measured values was compared. RESULTS No significant differences were found in the quantitative results of chlorogenic acid, emodin, chrysophanol, dl-tetrahydropalmatine determined by using QAMS and external standard method. CONCLUSION The QAMS method was reliable and accurate, which might be used for the quality control of Fule granula.
