Separation of Isomer and Similar Structure Compounds in Belamcandae Rhizoma by Methyl-β-CD Chiral Mobile Phase Additive HPLC

Abstract
OBJECTIVE To establish the separation method of isomer and similar compounds in Belamcandae Rhizoma with RP-HPLC. METHODS The separation was performed on Kromasil C18 column (250 mm×4.6 mm, 5 μm) with mobile phase of aqueous phase with 0.5% methyl-β-cyclodextrin and 0.1% phosphate, organic phase for acetonitrile, gradient elution. The column temperature was 30℃, and the detection wavelength was 265 nm. RESULTS Successful split isomer of iristectorigenin A with iristectorigenin B, with similar structure of irigenin. And it was possible to measure 8 compounds in Belamcandae Rhizoma at the same time. CONCLUSION Belamcandae Rhizoma can be baseline-separated by this method which is simple and economic with good resolution.
