Effects of Xuebijing Injection on Oxidative Stress and Early Inflammatory Factors in Rats with Sepsis Induced Liver Injury

Abstract
OBJECTIVE To observe the effect of Xuebijing injection in rats with sepsis induced liver injury and explain the mechanism from anti-oxidative stress and early inflammatory factors. METHODS Fifty-four healthy Wistar rats were randomly divided into control group, sepsis group and Xuebijing group. The rat model of sepsis was established with cecal ligation puncture (CLP). Serum samples and indices of liver tissue were measured at the point of 6, 12, 24 h after model was established. Observed the effect of Xuebijing injection on liver anti-oxidation, hapatocyte membrance permeability, and change of inflammatory cytokines. RESULTS Compared with control gruop, the concentrations of peroxidation in sepsis group and Xuebijing group were no significant difference after 6, 12 h of established rat model, however C-react protein(CRP) was much higher. After 12 h of CLP, MDA and TNF-α decreased and IL-10 began to increase in Xuebijing group compared with sepsis group. But ALT and AST in Xuebijing group was lower (P<0.05) than sepsis group after 24 h. The expression of GSH and SOD in livers of Xuebijing group were significantly higher (P<0.05), and MDA lower than sepsis group(P<0.05). Meanwhile, CRP, serum tumor TNF-α was decreased (P<0.01) and interleukin-10(IL-10) increased in Xuebijing group compared to sepsis group. According to liver histopathological changes, Xuebijing group showed better pathologic changes compaired to sepsis group after 12, 24 h(P < 0.05, P < 0.01). CONCLUSION Xuebijing injection show protective effect on acute liver injury of sepsis. Its mechanism may be correlated with anti-oxidative stress of liver, protection of hapatocyte membrance permeability and inhibition of some inflammatory factors.
