败酱总黄酮对局灶性脑缺血再灌注大鼠氧自由基及能量代谢的影响

摘要:
目的 观察败酱总黄酮对局灶性脑缺血再灌注大鼠氧化应激水平及能量代谢的影响。方法 将98只Wistar大鼠随机分为假手术组,模型组,尼莫地平组(20 mg·kg-1),脑络通组(500 mg·kg-1),败酱总黄酮高、中、低剂量组(200,100,50 mg·kg-1)。灌胃体积为20 mL·kg-1,每天给药1次,连续给药7 d。采用线栓法制作大鼠大脑中动脉闭塞模型。在模型制备前7 d给大鼠服用药物,连续7 d灌胃给药。再灌注22 h后进行神经功能评分,并测定脑组织中ATP酶、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平; HE染色观察脑部海马区及皮质区组织病理变化情况。结果 与假手术组相比,模型组大鼠神经功能缺失评分显著升高,Na+-K+-ATP酶和Mg2+-ATP酶活力均显著降低,脑匀浆中MDA含量显著升高、SOD含量显著降低(P<0.01)。与模型组相比,败酱总黄酮各剂量组可以显著降低脑缺血再灌注大鼠的神经功能评分,升高脑组织中SOD、ATP酶水平,降低MDA水平(P<0.01或P<0.05),改善脑组织中海马、皮质区的病理损伤。结论 败酱总黄酮对局灶性脑缺血再灌注损伤有较好的保护作用,可改善氧化应激水平,改善大脑皮质及海马区的病变情况,其作用机制可能与改善脑组织能量代谢,降低自由基损伤有关。

Effects of Total Flavonoids from Patriniae on Oxygen Free Radicals and Energy Metabolism in Rats with Focal Cerebral Ischemia Reperfusion

Abstract:
OBJECTIVE To observe the effects of total flavonoids from Patrinia on oxidative stress and energy metabolism in rats with focal cerebral ischemia reperfusion injury. METHODS Ninety-eight Wistar rats were randomly divided into sham operation group, model group, nimodipine group(20 mg·kg-1), Naoluotong group(500 mg·kg-1), high-dose, medium-dose, low-dose (200, 100, 50 mg·kg-1) Patrinia total flavonoids groups. Stomach irrigation volume was 20 mL·kg-1, once a day for 7 d. Middle cerebral artery occlusion models of rats were made with thread blocking method. Drugs were given to the rats 7 d before preparation of the models, ig for consecutive 7 d. After 22 hours of reperfusion, the neurological function score was measured, and the levels of ATPase, superoxide dismutase(SOD) and malondialdehyde(MDA) in brain tissue were measured. The histopathological changes of hippocampus and cortex were observed by HE staining. RESULTS Compared with the sham operation group, the neurological deficit score in the model group increased significantly, Na+-K+-ATP and Mg2+-ATP enzyme activities decreased significantly, MDA content in the brain homogenate increased significantly, and SOD content decreased significantly(P<0.01). Compared with the model group, total flavones of Patrinia could decrease the scores of neurological deficit, increase the levels of SOD and ATP in brain tissue, decrease MDA levels(P<0.01 or P<0.05), and improve the pathological damage of hippocampus and cortex in brain tissue. CONCLUSION Total flavones from Patrinia have better protective effect on focal cerebral ischemia-reperfusion injury. It can improve the level of oxidative stress and could improve the pathological changes of cerebral cortex and hippocampus. The mechanism of action may be related to improving energy metabolism of brain tissue and reducing free radical injury.
