HPLC测定酮缬氨酸钙含量的方法优化

摘要:
目的 优化HPLC测定酮缬氨酸钙含量的方法。方法 HPLC采用CAPCELL PAK C18 MGⅡ柱(250 mmx4.6 mm,5 μm),流动相为高氯酸钠溶液(pH 2.5)-乙腈(96∶4),柱温为35℃,检测波长205 nm,流速1.0 mL·min-1结果 酮缬氨酸钙在0.025~0.185 mg·mL-1内有良好的线性关系(r=1.000 0),平均回收率(n=9)为100.0%。结论 该方法简便、快速、准确,灵敏度高,重复性好,可用于酮缬氨酸钙的含量测定。

Abstract:
OBJECTIVE To improve an HPLC method for the content determination of α-ketovaine calcium. METHODS The HPLC method was performed on CAPCELL PAK C18 MGⅡ column(250 mm×4.6 mm, 5 μm) with mobile phase of sodium perchlorate buffer(pH 2.5)-acetonitrile(96:4). Column temperature was 35℃, detective wavelength was 205 nm, and the flow rate was 1.0 mL·min-1. RESULTS The linear range of α-ketovaine calcium was 0.025-0.185 mg·mL-1 (r=1.000 0), the average recovery(n=9) was 100.0%. CONCLUSION The method is simple, rapid and accurate with good repeatability, which can be used for the determination of α-ketovaine calcium.
