秦巴硒菇提取物(FA-2-b-β)对慢性髓系白血病原代细胞凋亡的影响

摘要:
目的 探讨中药秦巴硒菇提取物(酸性RNA蛋白复合物FA-2-b-β)对人慢性髓系白血病(chronic myeloid leukemia,CML)原代细胞系凋亡的影响。方法 采用CCK8法检测不同时间段(24,48,72 h)FA-2-b-β作用于CML原代细胞的增殖抑制率;流式细胞术检测细胞凋亡率和周期;蛋白印迹法测定相关凋亡蛋白Bcl-2、Bax、MET、CD44和β-catenin的表达。结果 FA-2-b-β抑制CML原代细胞增殖,并呈时间、浓度依赖性。FA-2-b-β处理后,CML原代细胞凋亡率明显升高,促凋亡蛋白Bax表达升高,抗凋亡蛋白Bcl-2、MET、CD44和β-catenin表达降低。结论 秦巴硒菇提取物通过下调凋亡相关蛋白抑制CML原代细胞增殖并诱导细胞凋亡,表现出较强的体外抗白血病作用。

Abstract:
OBJECTIVE To investigate the effect of acidic RNA protein complex (FA-2-b-β) extracted from Qinba mushroom on apoptosis of primary human chronic myeloid leukemia(CML) bone marrow cell line. METHODS The cell proliferation rate was measured by CCK8 method at different time(24, 48, 72 h) points. Flow cytometry was used to detect cell apoptotic rate and cell cycle. Western blotting was used to detect the expression of Bcl-2, Bax, MET, CD44 and β-catenin protein. RESULTS FA-2-b-β inhibited the proliferation of primary CML bone marrow cell line effectively in a dose and time dependent manner. After treated with Qinba mushroom, the apoptosis rates of primary CML bone marrow cell line increased, the expression of Bax increased, and the expression of Bcl-2, MET, CD44, β-catenin decreased. CONCLUSION Qinba mushroom extract inhibits proliferation, induces apoptosis of primary CML bone marrow cell line by regulating the related apoptotic protein. Qinba mushroom has strong anti-leukemia effect in vitro.
