白藜芦醇对长期高热量饮食小鼠的认知功能保护及突触相关机制研究

摘要:
目的 观察白藜芦醇(resveratrol,RES)对长期高热量饮食(high-calorie diet,HCD)小鼠认知功能的影响并探讨其相关机制。方法 50只C57BL/6小鼠随机均分为对照组、HCD组、HCD+25 mg·kg-1RES组(低剂量组)、HCD+50 mg·kg-1RES组(中剂量组)和HCD+100 mg·kg-1RES组(高剂量组)。6个月后,检测小鼠基本生理指标,并利用Morris水迷宫和条件恐惧试验检测认知水平,RT-PCR和ELISA分别检测海马BDNF、Trk B、SYP和PSD95 mRNA及蛋白水平。结果 与HCD组比较,RES干预使小鼠体质量、肥胖指数和脂肪系数均明显降低,空腹血糖、胰岛素和HOMA-IR明显降低,海马依赖性学习记忆明显提高,海马组织BDNF、Trk B、SYP和PSD95 mRNA和蛋白水平明显升高,以高剂量组更显著。结论 RES可通过上调海马BDNF、Trk B、SYP、PSD95基因和蛋白表达改善HCD小鼠学习记忆能力。

Abstract:
OBJECTIVE To explore the protective effect of resveratrol(RES) on cognitive function of mice fed long-term high-calorie diet(HCD) and its synapse-related mechanisms. METHODS Fifty male C57BL/6 mice were randomly divided equally into control group, HCD group, HCD+25 mg·kg-1RES group(low dose group), HCD+50 mg·kg-1 RES group(middle dose group) and HCD+100 mg·kg-1 RES(high dose group). Six months later, the basic physiological indexes of mice were measured. The cognitive level was measured by Morris water maze and conditional fear test. And the mRNA and protein levels of BDNF, Trk B, SYP and PSD95 in hippocampus were detected by RT-PCR and ELISA, respectively. RESULTS Compared with HCD group, the RES groups demonstrated decreased the body mass, obesity index and fat index of mice, decreased the value of fasting blood glucose and insulin, and HOMA-IR, improved the level of hippocampus-dependent learning and memory, increased the mRNA and protein levels of BDNF, Trk B, SYP and PSD95 in hippocampus, especially for the high dose group. CONCLUSION RES can improve the learning and memory ability of HCD mice by up-regulating the expression of BDNF, Trk B, SYP and PSD95 in hippocampus.
