丹红注射液长期给药对大鼠血管舒张功能的影响

摘要:
目的 探索丹红注射液(Danhong injection,DHI)长期给药对正常大鼠血管舒张功能的影响。方法 20只正常SD大鼠随机分为2组,DHI给药组和空白组,连续静脉注射给药28 d后,乙酰胆碱(ACh)和硝普钠(SNP)分别检测DHI对大鼠胸主动脉的舒张功能的影响。Griess reagent、ELISA试剂盒分别检测大鼠血清中一氧化氮(NO)、前列环素(epoprostenol,PGI2)的含量变化。实时定量RT-PCR检测动脉血管间ACs、cAMP以及PKAs基因表达变化。结果 与空白组比较,DHI长期给药后能够显著性增加ACh浓度依赖性的血管舒张功能,而不改变SNP浓度依赖性的血管舒张功能;显著增加大鼠血清中PGI2浓度而不改变NO浓度;RT-PCR显示,DHI组的ACs、cAMP以及PKAs基因表达变化分别上调约1.21,1.78以及1.48倍。结论 DHI长期给药后能显著改善大鼠内皮依赖性的血管舒张功能,其作用机制很大程度是通过干扰PGI2介导的ACs/cAMP/PKAs信号途径实现的。

Vasodilative Effect of Danhong Injection Long-term Treatment in Rats

Abstract:
OBJECTIVE To investigate long-term treatment effect of Danhong injection(DHI) on vasodilation in normal rats. METHODS Twenty normal male SD rats were divided into two groups randomly, DHI group and control group. We performed continuous intravenous administration during 28 days, and diastolic function of DHI in rats was detected by Ach and SNP. By applying Griess reagent kit/ELISA kit, checked the serum contents of nitric oxide(NO)/epoprostenol(PGI2) in rats. Additionally, real time PCR(RT-PCR) was carried out to monitor the gene expression change of ACs, cAMP and PKAs in each group. RESULTS Compared with control group, DHI long-term treatment could significantly increase rats' vasodilation in response to ACh stimulation. However, it didn't exhibit close association with SNP stimulation. Moreover, DHI long-term treatment would increase PGI2 level in serum without an impact on NO in serum. Meanwhile, RT-PCR showed that the expression of ACs, cAMP and PKAs in DHI group were up-regulated by 1.21, 1.78 and 1.48 times, respectively. CONCLUSION DHI long-term treatment can significantly improve endothelial-dependent vasodilation in rats by interfering ACs/cAMP/PKAs signal pathway mediated by PGI2.
