疏肝健脾活血方对肝郁脾虚证FD大鼠血浆脑肠肽的影响

摘要:
目的 观察疏肝健脾活血方对肝郁脾虚证功能性消化不良(functional dyspepsia,FD)大鼠血浆脑肠肽的影响。方法 70只SPF级♀SD大鼠按体质量随机抽取10只作为正常组,其余60只采用“慢性束缚+过度疲劳+饮食失节”复合因素法造模21 d,造模成功后,按体质量随机分为模型组、疏肝健脾活血方高剂量组、中剂量组、低剂量组、柴芍六君子汤组、多潘立酮组。分组后,大鼠灌胃给予相应药物干预14 d,正常组和模型组大鼠灌胃给予相应体积纯净水。采用ELISA法检测大鼠血浆脑肠肽。结果 与模型组比较,疏肝健脾活血方各组大鼠血浆胃促生长素(Ghrelin)、胃动素(motilin,MTL)含量显著增加(P<0.05),胆囊收缩素(Cholecystokinin,CCK)、降钙素基因相关肽(Calcitonin gene related peptide,CGRP)含量显著降低(P<0.01),高剂量组大鼠血浆胃泌素(gastrin,GAS)含量显著增加(P<0.05),疏肝健脾活血方高剂量组、中剂量组大鼠血浆血管活性肠肽(vasoactive intestinal peptide,VIP)含量显著降低(P<0.05)。结论 疏肝健脾活血方具有调节肝郁脾虚证FD大鼠Ghrelin、MTL、CCK、VIP、GAS、CGRP的作用。

Abstract:
OBJECTIVE To observe the effect of Shugan Jianpi Huoxue(SGJPHX) decoction on brain-gut peptides in plasma of functional dyspepsia(FD) rats with liver depression and spleen deficiency. METHODS Seventy SPF female rats were randomly divided into normal group(10) and modeling group(60) according to the body weight of rats. The modeling group rats were adopted by chronic restraint stress+excess fatigue+improper diet for 21 d to form the model. After modeling, the modeling group rats were randomly divided into model group, SGJPHX high dose group, medium dose group, low dose group, Chaishao Liujunzi(CSLJZ) decoction group and domperidone group. After group division, the SGJPHX high dose group, medium dose group and low dose group rats were given SGJPHX decoction, the CSLJZ group rats were given CSLJZ decoction, the domperidone group rats were given domperidone, and the model group and normal group were given pure water for 14 d. The brain-gut peptides in plasma of FD Rats were detected by ELISA. RESULTS Compared with the model group, the content of Ghrelin, MTL in plasma of SGJPHX each dose group were significantly increased(P<0.05), and the CCK, CGRP were significantly reduced(P<0.01). The GAS in plasma of SGJPHX high dose group was significantly increased(P<0.05), the VIP in plasma of SGJPHX high dose group and medium dose group were significantly reduced(P<0.05). CONCLUSION SGJPHX decoction can regulate brain-gut peptides in plasma of FD rats with liver depression and spleen deficiency.
