虎杖苷对去卵巢骨质疏松大鼠β-Catenin及GSK-3β蛋白表达的影响

摘要:
目的 研究虎杖苷对卵巢摘除骨质疏松大鼠β-Catenin及GSK-3β蛋白表达的影响,探索其对卵巢摘除骨质疏松大鼠的保护作用及可能的作用机制。方法 SPF级♀大鼠60只,随机分为假手术组、模型组、虎杖苷低(5 mg·kg-1·d-1)、中(15 mg·kg-1·d-1)、高剂量组(45 mg·kg-1·d-1),每组12只,灌胃给药。除假手术组外,其余各组采用摘除大鼠双侧卵巢的方法模拟骨质疏松模型。12周后,用双能X线吸收法测量骨密度(bone mineral density,BMD),ELISA法检测E2、ALP、TRAP等生化指标的水平;western blotting法检测β-Catenin和GSK-3β蛋白的表达,qRTPCR法检测β-Catenin和GSK-3β mRNA的表达。结果 与模型组比较,虎杖苷各组BMD,E2显著升高(P>0.01)、血清ALP、TRAP显著降低(P>0.01);β-Catenin蛋白的表达升高(P>0.01),GSK-3β表达降低(P>0.01);β-Catenin mRNA表达显著升高(P>0.01),GSK-3β mRNA表达显著降低(P>0.01)。结论 虎杖苷对去卵巢骨质疏松有一定的治疗作用,其机制可能与增加β-Catenin表达,减少GSK-3b表达有关。

Effect of Polydatin on the Expression of Wnt/β-Catenin signal pathway in ovariectomized osteoporosis rats

Abstract:
OBJECTIVE To study the effect of polydatin(PD) on the expression of β-Catenin and GSK-3β protein in ovariectomy osteoporosis rats. METHODS The 60 female SPF rats were randomly divided into sham operation group, model group, PD low(5 mg·kg-1·d-1), middle(15 mg·kg-1·d-1) and high dose(45 mg·kg-1·d-1) groups (12 rats in each group, ig), every group were established by ovariectomy except sham group. After 12 weeks, the bone mineral density (BMD) of these rats was detected by dual energy X-ray absorptionmetry, ELISA method was used to detect serum estradiol(E2), bone alkaline phosphatase(ALP), tartrate resistant acid phosphatase(TRAP) and other biochemical indicators of the level; western blotting was used to detect the expression of β-Catenin and GSK-3β protein; qRTPCR was used to detect the expression of β-Catenin and GSK-3β mRNA. RESULTS Compared with model group, PD groups increased BMD, E2 significantly(P>0.01), decreased ALP, TRAP level(P>0.01) and increased the expression of β-Catenin protein and mRNA(P>0.01); decreased the expression of GSK-3β protein and mRNA (P>0.01). CONCLUSION PD has therapy effects in ovariectomy osteoporosis rats. Its mechanism could be relative with increasing the expression of β-Catenin protein and decreasing expression of GSK-3β.
