基于HPLC多波长融合指纹图谱及谱效关系的熟地黄活性成分研究

摘要:
目的 建立熟地黄HPLC多波长融合指纹图谱并筛选抗糖尿病骨质疏松的活性成分。方法 采用HPLC建立10批熟地黄多波长(203,210,254,334 nm)融合指纹图谱;通过糖尿病骨质疏松大鼠模型药效评价,运用偏最小二乘回归法进行谱效分析,得到主要活性成分并以高糖成骨细胞模型验证其药效,并考察熟地黄的体内抗糖尿病骨质疏松作用。结果 10批熟地黄融合指纹图谱共确定26个共有峰,鉴定出的17个色谱峰与骨形成指标(血清碱性磷酸酶活性)呈正相关,其中峰8(梓醇)和峰12(地黄苷D)的标准回归系数和VIP值最显著,二者均能提高成骨细胞增殖活性和细胞碱性磷酸酶水平。结论 梓醇和地黄苷D具有显著抗糖尿病骨质疏松作用,为熟地黄主要活性成分。多波长融合指纹图谱结合谱效关系可作为活性成分筛选的有效策略。

Study on Active Constituents of Rehmanniae Radix Praeparata Based on HPLC Multi Wavelength Fusion Fingerprint and Spectral Effect Relationship

Abstract:
OBJECTIVE To establish HPLC multi-wavelength fusion fingerprints of Rehmanniae Radix Praeparata and to screen the anti-diabetic osteoporosis active ingredients. METHODS The multi-wavelength(203, 210, 254, 334 nm) fusion fingerprints of 10 batches of Rehmanniae Radix Praeparata were established by HPLC. Through the efficacy evaluation of the diabetic osteoporosis rat model, the partial least squares regression method was used, the main active components were obtained and the efficacy was verified by the high glucose osteoblasts model, and the anti-diabetic osteoporosis effect of Rehmanniae Radix Praeparata was investigated. RESULTS A total of 26 common peaks were identified in the fusion fingerprints of 10 batches of Rehmanniae Radix Praeparata, and 17 peaks were positively correlated with the bone formation marker(serum alkaline phosphatase activity). Regression coefficients and VIP values of peak 8(catalpol) and peak 12(rehmannioside D) were the most significant. Both osteoblast proliferation activity and cellular alkaline phosphatase levels. CONCLUSION Catalpol and rehmannioside D have significant anti-diabetic osteoporotic effect, and might be the main active components of Rehmanniae Radix Praeparata. The multi-wavelength fusion fingerprint combined with the spectral effect relationship can be used as an effective strategy for the screening of active ingredients.
