玉竹多糖对2型糖尿病大鼠糖脂代谢的影响及机制研究

摘要:
目的 研究玉竹多糖对2型糖尿病大鼠糖脂代谢的影响以及作用机制。方法 注射链脲佐菌素-枸橼酸钠溶液构建2型糖尿病大鼠模型。考察玉竹多糖对糖尿病大鼠的生理指标、血液指标和生化指标的影响;HE染色和O油红染色观察大鼠肝脏细胞的病理改变情况。Western blotting法验证大鼠糖脂代谢的作用机制。结果 玉竹多糖能够显著降低模型大鼠的谷丙转氨酶、谷草转氨酶、总胆固醇、甘油三酯、低密度脂蛋白、肌酐、血尿素氮、糖化血红蛋白、丙二醛水平,显著提高其高密度脂蛋白、C肽、胰岛素、超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶水平;在病理水平上,玉竹多糖能够明显改善模型大鼠肝脏细胞病变程度。Western blotting结果显示玉竹多糖能够上调Nrf2介导的HO-1参与PI3K/AKT途径调节细胞脂代谢过程。结论 玉竹多糖能够保护机体免受链脲佐菌素诱导的病变过程,对2型糖尿病的治疗具有积极作用。

Abstract:
OBJECTIVE To study the effect of polygonatum polysaccharide on glucose and lipid metabolism in type 2 diabetic rats and its mechanism.METHODS The type 2 diabetic rat model was constructed by injecting streptozotocin-sodium citrate solution to explore the effects of polygonatum polysaccharide on the physiological, blood and biochemical indicators of rats.The pathological changes of rat liver cells were observed by HE staining and oil red O staining.Western blotting was used to verify the mechanism of glucose and lipid metabolism in rats.RESULTS Polygonatum polysaccharide could significantly reduce alanine transaminase, aspartate transaminase, total cholesterol, triglyceride, low density lipoprotein cholesterol, creatinine, blood urea nitrogen, glycosylated hemoglobin and malondialdehyde, significantly increased their high density lipoprotein cholesterol, C-peptide, insulin, superoxide dismutase, catalase and glutathione peroxidase, and was able to improve the pathological degree of hepatocyte in rats.Western blotting results showed that polygonatum polysaccharide could upregulate the process of cell lipid metabolism through the PI3K/Akt pathway.CONCLUSION Polygonatum polysaccharide can protect the body from the pathological process induced by streptozotocin, and has a positive effect on the treatment of type 2 diabetes.
