促排合剂提取工艺研究

摘要:
目的 优选促排合剂的提取工艺。方法 采用正交设计试验对促排合剂提取工艺的影响因素进行设计,并在设计的工艺条件下进行试验,以处方中有效成分2, 3, 5, 4-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量及出膏率为考察指标,筛选出促排合剂最佳提取工艺。结果 最佳工艺条件为加10倍于药材量的水,浸泡药材22 h,煎煮3次,每次煎煮2 h。对所选最优结果重复3次验证,促排合剂出膏率为61.23%~62.04%,有效成分2, 3, 4, 5-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量为0.48~0.58 mg·mL-1。结论 所优选的工艺条件合理、可行。

Abstract:
OBJECTIVE To optimize the extraction process for Cupai mixture by orthogonal design. METHODS Orthogonal design method L9(43) was applied to optimization of extraction process for Cupai mixture, and the contents of 2,3,5,4-tetrahydroxy toluylene-2-O-β-D-glucoside in the extract and the percentage of extract were used as indices for evaluation of the extraction process. RESULTS The optimal extraction process was characterized as follows. The water used was 10 times the amount of crude drugs. The crude drug should be immersed for 22 h and decoction should be repeated for three times, each of which lasted for 2 h. The results of optimized extraction process were validated for three times. The mean value for the extract percentage were 61.23%-62.04%, and the effective component 2,3,5,4-tetrahydroxy toluylene-2-O-β-D-glucoside were 0.48-0.58 mg·mL-1. CONCLUSION The optimized extraction method is reasonable and feasible.
