2型糖尿病大血管病变与内脂素的关系探讨及降糖调脂灵的干预作用

    Preventive Effect of Jiangtang Tiaozhiling on T2DM Macroangiopathy and Role of Visfatin Expression

    • 摘要: 目的 观察降糖调脂灵保护2型糖尿病大血管病变与内脂素的关系。方法 10只健康成年♂Wistar大鼠作为正常对照组,喂以标准饲料不做处理;50只健康♂大鼠通过高糖高脂饲料喂养+腹腔注射链脲佐菌素诱发糖尿病,随机选取30只成模大鼠分为3组:糖尿病模型组(n=10,灌胃,等量生理盐水)、二甲双胍组(n=10,灌胃,二甲双胍200 mg·kg-1·d-1)、降糖调脂灵组(n=10,灌胃,降糖调脂灵浓缩液5 mL·kg-1·d-1),继续喂以高糖高脂饲料。12周后取血测血糖、血脂、单核细胞趋化因子和细胞间黏附分子的含量;Western-blot法检测血清内脂素表达水平;光镜观察主动脉壁情况,透射电镜观察主动脉超微结构改变。结果 降糖调脂灵组血糖、血脂、单核细胞趋化因子和细胞间黏附分子的含量降低,内脂素表达减少,主动脉内皮细胞损坏减轻。结论 降糖调脂灵保护T2DM大血管病变,可能与抑制内脂素表达,降低血糖、血脂,减轻炎症反应有关。

       

      Abstract: OBJECTIVE To observe the preventive effect of Jiangtang Tiaozhiling on T2DM angiopathy and the role of visfatin expression. METHODS Ten health male Wistar rats in normal control group were only fed with standard diet. Fifty health male Wistar rats were fed with high glucose and fat diet+injected STZ intraperitoneal to induce diabetes mellitus. Thirty diabetes mellitus rats were randomly divided into 3 groups: diabetes mellitus model group(n=10, intragastric administration, td NS); Metformin group(n=10, intragastric administration, metformin 200 mg·kg-1·d-1) and Jiangtang Tiaozhiling group(n=10, intragastric administration, Jiangtang Tiaozhiling concentrated solution 5 mL·kg-1·d-1) and fed with high glucose and fat diet. After 12 weeks, serum glucose, lipids, serum level of monocyte chemotactic factor and intercellular adhesion molecule were checked; the expression of visfatin was detcted by Western-blot method, the aorta thoracalis wall was observed by light microscope and the ultramicrostructural of aorta thoracalis endothelial cell was observed by electron microscope. RESULTS In Jiangtang Tiaozhiling group, the levels of serum glucose, lipids, monocyte chemotactic factor and intercellular adhesion molecule were significant decreased; the expression of visfatin diminished and aorta thoracalis endothelial cell damaged slightly. CONCLUSION Jiangtang Tiaozhiling protects T2DM macroangiopathy, its mechanism may be related to inhibition expression of visfatin and decrease serum glucose, blood lipids level, attenuated inflammatory reaction.
