PTD-SOD对Na2S2O4诱导的SH-SY5Y细胞损伤的保护和修复作用研究

    Protective Effects of PTD-SOD Fusion Protein on SH-SY5Y Cell Induced by Sodium Dithionite Sodium Hydrosulfite

    • 摘要: 目的 研究PTD-SOD对Na2S2O4诱导的SH-SY5Y细胞损伤的保护和修复作用。方法 建立Na2S2O4诱导的SH-SY5Y细胞缺氧损伤模型,采用MTT法检测融合蛋白PTD-SOD对细胞存活率的影响,LDH释放分析检测PTD-SOD对细胞损伤后的保护作用及损伤后细胞内氧化与抗氧化水平的变化。结果 对Na2S2O4诱导的SH-SY5Y细胞缺氧损伤,MTT活性检测、LDH分析及氧化与抗氧化水平分析都表明PTD-SOD能明显提高细胞存活率和细胞内抗氧化能力。结论 PTD-SOD融合蛋白能保护Na2S2O4诱导缺氧损伤的SH-SY5Y细胞,具有抑制细胞死亡,改善损伤细胞内抗氧化能力。

       

      Abstract: OBJECTIVE To investigate the protection mechanism of fusion protein PTD-SOD on SH-SY5Y cell against toxicity induced by sodium dithionite sodium hydrosulfite. METHODS Establish the hypoxia damage model of SH-SY5Y cell induced by Na2S2O4. MTT assay was used to measure the survival of SH-SY5Y cell. LDH, SOD and MDA were measured by spectrophotometric method to analysis the protective effect of PTD-SOD on SH-SY5Y cell and the change of the cell oxidation and antioxidant levels after the hypoxia damage. RESULTS MTT assay demonstrated that PTD-SOD could promote significantly the survival of the cell. LDH release assay and SOD and MDA assay demonstrated that PTD-SOD could promote the levels of antioxidant against the hypoxia damage. CONCLUSION The fusion protein PTD-SOD protects SH-SY5Y cell from death after injury induced by Na2S2O4 and promotes antioxidant ability in cell.
